abstract | The invention discloses a ruthenium-catalyzed ortho-meta selective hydrogen-deuterium exchange reaction of a benzene ring. Specifically, the invention provides a preparation method of a deuterium-labeled compound shown as a formula I, which comprises the following steps: under the catalysis of dichlorobis (4-methyl isopropylphenyl) ruthenium, a compound shown as a formula II reacts with deuterated acetic acid in the presence of silver acetate and triphenylphosphine to obtain a deuterium-labeled compound shown as a formula I. The invention adopts dichlorobis (4-methyl isopropyl phenyl) ruthenium as a catalyst, widens the application range of a guide group, is not limited to groups such as pyrazole, pyridine and the like, such as other nitrogen heterocyclic aromatic rings, for example: pyrazine, pyrimidine, benzimidazole and the like can well induce the high-efficiency hydrogen and deuterium exchange of ortho-position and meta-position of a benzene ring, and the highest deuterium substitution rate can reach 95%. The reaction of the invention can simultaneously realize ortho-meta deuteration of benzene ring, can introduce a plurality of deuterium atoms, provides another simple and efficient method for synthesis of internal standard reagent, and has higher social benefit and economic benefit. |
