abstract | The invention provides an enrichment and identification method of an RNA-protein complex. The PB reagent is shown as a formula I, and can perform covalent reaction with pyrimidine bases of RNA in the RNA-protein complex under 365nm wavelength ultraviolet light, so that specific enrichment of the RNA-protein complex is realized. The enrichment method has the following advantages: (1) the method can be used for unbiased marking and enrichment of various types of RNA and protein complexes thereof, and especially has important significance on the enrichment identification of non-coding RNA-protein complexes in the field of epigenetic research; (2) PB reagent enrichment does not need metabolic labeling, so that the normal physiological state of cells is less disturbed, and the labeling efficiency is higher; (3) the kit can be applied to the comprehensive research of various RNA-protein complexes in tissue samples, and (4) the ethylene glycol repeating unit is used as a connecting arm for connecting psoralen and biotin in a PB reagent, so that the steric hindrance can be effectively reduced, and the enrichment efficiency of streptomycin modified magnetic beads on the RNA-protein complexes can be improved. |
