abstract The invention discloses a construction method of enterotoxin gene LT knocked-out escherichia coli, which comprises the following steps: 1) constructing LT gene homologous recombination donor LT donar, wherein the base sequence of LT donar sequentially comprises L-arm, cat promoter, CmR, sgCommon, PAM, L-short and R-arm polynucleotide fragments; and 2) preparing competent cells containing pCas-Red plasmids, adding the LT donar in the step 1) for transformation, and then screening positive clones to obtain the Escherichia coli with the heat-labile enterotoxin gene LT knocked out tracelessly. According to the invention, Crispr/Cas9 is combined with a lambda-Red homologous recombination system, the LT gene of enterotoxigenic Escherichia coli K88 is successfully subjected to traceless knockout, a mutant strain loses hemolytic ability, and growth is slowed down; the method can quickly and conveniently knock out enterotoxigenic Escherichia coli K88 gene without trace, and provides a solid research foundation for researching LT function.
