abstract The invention discloses an ethanol detection method and a kit. The method utilizes that when ethanol is oxidized by alcohol dehydrogenase to generate acetaldehyde, oxidized coenzyme I generates reduced coenzyme I, and the generated reduced coenzyme I converts potassium ferricyanide into potassium ferrocyanide, potassium ferrocyanide Prussian blue is produced with ferric chloride, and the amount of Prussian blue produced is determined by measuring the absorbance of Prussian blue, and the ethanol content is calculated. The kit of the invention consists of a mixed solution of 0.2-2U alcohol dehydrogenase, 0.2-2U NADH dehydrogenase, 0.2-2mM oxidized coenzyme I and 10-100mM potassium ferricyanide, buffer and 10-100mM trichloride iron. The invention can measure the ethanol concentration in the liquid in a short time only by using a microplate reader that only supports the wavelength of visible light, and the detection limit is as low as 0.1 mM.
