abstract | The invention relates to a method for synthesizing head-tail cyclic peptide containing alanine, which comprises the following steps: 1) selecting solid-phase synthetic resin; 2) firstly, Fmoc-Asp (OH) -OAll is coupled with solid-phase synthetic resin, then NH2-Asp- (solid-phase synthetic resin) -OAll is obtained according to Fmoc/tBu solid-phase synthetic strategy, and amino acid residues in a peptide sequence are coupled in sequence to obtain NH2-AAn-Asp- (solid-phase synthetic resin) -OAll; 3) removing a protecting group All from a solid phase to obtain NH2-AAn-Asp- (solid phase synthetic resin) -OH; 4) under the solid phase condition, carrying out head-tail condensation cyclization on head-tail amino acid of the polypeptide long chain obtained in the step 3); 5) cracking the product obtained in the step 4) to obtain solid-phase synthetic resin, and preparing intermediate cyclic peptide crude peptide; 6) and (3) removing side chain carboxyl of terminal aspartic acid from the intermediate cyclopeptide crude peptide under the action of decarboxylase to obtain a finished product. |
