The method relates to five-step reaction, which comprises the steps of firstly obtaining a key intermediate 6 through nucleophilic substitution, hydrolysis and silicon etherification, and then obtaining a final product PA-824 through O-alkylation and cyclization reactions. The raw materials needed by the whole reaction are cheap and easy to obtain, the use of explosive raw material 2, 4-dinitroimidazole is avoided, the reaction condition is mild, the operation is simple, and the large-scale production is easy.
