abstract | The invention discloses a method for screening the radiation mutagenic dose of iris. By constructing the tissue-cultured seedlings of the iris clones, the tissue-cultured seedlings of the iris clones are subjected to radiation mutagenesis treatment with different doses. The tissue cultured seedlings were treated for 40 days, and the malondialdehyde (MDA) content, superoxide anion free radical (O 2 -· ) production rate, H 2 O 2 content, and antioxidant enzyme (SOD, CAT) activity were measured. MDA and H 2 O 2 content, O 2 -· production rate, SOD and CAT activity at 40 days after mutagenesis treatment were used to determine the appropriate mutagen dose. The less appropriate the mutagenic dose, the smaller the measured value and the higher the survival rate, the more appropriate the corresponding radiation mutagenic dose. The present invention firstly constructs tissue-cultured seedlings of iris clones as mutagenesis materials, which overcomes the problem of separation of certain traits when hybrid offspring seeds are used as mutagenesis materials. Correlation analysis, screening out the physiological and biochemical indicators related to the survival rate of the mutagenic line, used to screen the appropriate mutagenic dose of iris. |
