abstract | The invention discloses an extraction, purification and identification method of moso bamboo shoot high-activity mitochondria. The invention provides a method for extracting moso bamboo shoot mitochondria, which comprises the following steps: homogenizing moso bamboo shoots, performing differential centrifugation and sucrose density gradient centrifugation in sequence to obtain purified mitochondria; the centrifugal force adopted by the differential centrifugation is 1300-1700g, 5000-7000g and 12000-15000g in sequence. The method has the advantages of small damage to mitochondria, high activity of extracted mitochondria, thorough separation of nucleus components, chloroplast or proplast components, phenols, polysaccharides and the like outside the mitochondria, and extraction of high-quality mitochondrial RNA from purified mitochondria, which lays a good foundation for subsequent molecular biology experiments, such as PCR reaction, cDNA library construction, mitochondrial transcriptome research and the like. |
