abstract |
Provided herein are methods that enable parallel assessment of multiple functional nucleic acids in individual cells or subpopulations of cells in the context of incubation with other types of single cells. A key insight is the simultaneous measurement of polynucleotides derived from small populations of at least two different cell types, allowing the function of one cell type to be correlated with the clonal identity of another cell. These methods process thousands, millions or more single cells or small cell populations simultaneously. The method integrates molecular, algorithmic and engineering approaches. The present invention has broad and useful applications in many biological and medical fields, including immunology and drug discovery. |
