abstract | The present invention provides a method for capturing nucleic acid comprising a target oligonucleotide sequence from a nucleic acid library, comprising: contacting the nucleic acid library with a probe, the nucleic acid library comprising a nucleic acid comprising a target oligonucleotide sequence, the probe comprising a sequence complementary to a target oligonucleotide sequence, wherein the contacting may be performed in a tetramethylammonium chloride (TMAC) based buffer at a temperature of about 60°C to about 70°C, the contacting may be The target oligonucleotide sequence is hybridized to a sequence complementary to the target oligonucleotide sequence, thereby producing a hybridization product; the hybridization product is separated from nucleic acids in the library that do not contain the target oligonucleotide sequence.
