abstract | The invention provides a T cell capable of stably expressing CAR and a preparation method and application thereof. The preparation method of the T cell stably expressing the CAR comprises the following steps: the CAR gene targeting the cancer cell surface antigen is introduced into T cells via a non-viral plasmid such that the CAR is stably expressed for more than 3 weeks and the foreign gene is not integrated into the T cell chromosome. The invention provides a non-integrated T cell for stably expressing CAR by a non-viral plasmid and a preparation method thereof for the first time, and the special plasmid can be stably expressed for more than 3 weeks after being introduced into the T cell by an electrotransfer method. The invention can effectively avoid the adverse effect of lentivirus and retrovirus on the genome modification of T cells, and can avoid introducing any lentivirus or retrovirus element into the T cells. |
