abstract |
Provided herein are systems for inducible expression of chimeric antigen receptors in cells. The system comprises: a) a first nucleic acid comprising a first promoter inducible by an agent, wherein the first nucleic acid is operably linked to encode a chimeric antigen receptor (the chimeric antigen); receptor comprising a ligand binding domain), a second polynucleotide (the second polynucleotide encodes a spacer), a third polynucleotide (the third polynucleotide nucleotide encoding a transmembrane domain), and a fourth polynucleotide (the fourth polynucleotide encoding an intracellular signaling domain); and b) a second nucleic acid comprising the second a promoter operably linked to a nucleic acid encoding a transcriptional activator of the first promoter induced by the agent. Also provided are methods of making such cells and methods of using these cells for therapy. |
