abstract The invention relates to the field of cell model construction, in particular to a CRISPR/Cas9 gene editing method for knocking out PD-L1 gene in keratinocytes, a keratinocyte model constructed by the method, and applications thereof. The advantage of the present invention is that the PD-L1 knockout keratinocyte stable transfection strain constructed by the present invention can not only be used for the research of psoriasis, but also can be used for the research of other skin diseases with low PD-L1 expression. The construction of PD-L1 knockout cell lines will help to explore the role of PD-L1 in psoriasis, pityriasis rosea, allergic contact dermatitis and other skin-related diseases, and further explore the role of PD-L1 in keratinocytes function.
