abstract | The invention discloses a novel synthetic method of a Flindokalner racemate. 3-fluoro-3- (2, 2, 2-trifluoro-1, 1-dihydroxyethyl) -6 (trifluoromethyl) indoline-2-ketone is taken as a raw material, and bromo reaction is carried out to obtain monofluoro bromo-2-indolinone (compound (1)); reacting the compound (1) with benzyl halide to generate 1-benzyl-3-bromo-3-fluoro-6- (trifluoromethyl) indolin-2-one (compound (2)); carrying out Suzuki coupling on the compound (2) and 5-chloro-2-methoxyphenylboronic acid pinacol ester to obtain 1-benzyl-3- (5-chloro-2-methoxyphenyl) -3-fluoro-6- (trifluoromethyl) indolin-2-one (a compound (3)); the compound (3) is debenzylated to give racemic flindakolner (compound (4)). The synthesis method has the characteristics of easily obtained raw material reagents, short reaction steps, mild reaction conditions, simple and convenient operation and the like. |
