abstract The invention belongs to the technical field of enzyme genetic engineering, and particularly relates to a phospholipase D mutant and preparation and application thereof. The technical scheme includes that site-directed mutagenesis is carried out on a wild phospholipase D gene by means of a recombinant DNA technology to obtain a phospholipase D gene with high activity, then the high-activity phospholipase D gene is expressed in a bacillus subtilis expression system and a pichia pastoris expression system (including a pichia pastoris free expression system and a pichia pastoris cell surface display system), a recombinant strain for producing the high-activity phospholipase D is obtained, after expression, the specific enzyme activity of the high-activity phospholipase D is detected to be improved by 38-140% compared with that of the wild phospholipase D, and the highest values of the fermentation enzyme activities of the high-activity phospholipase D in the bacillus subtilis expression system, the pichia pastoris expression system and the pichia pastoris cell surface display system are respectively 36.8U/ml, 48.1U/ml and 140.2U/(g.cell dry weight).
