titleOfInvention Application of the transcription factor C/EBP α as the transcription factor of ACOX1 promoter region
abstract The present invention provides a kind of transcription factor C/EBP α application of the transcription factor as ACOX1 promoter region; specifically; 5 ACOX1 promoter deletion segments have been cloned from cow genome group DNA; it is connected to and constructs the Dual-Luciferase report carrier of 5 recombinations in Dual-Luciferase report carrier pGL3-Basic; respectively with C/EBP α over-express vector cotransfection; the fluorescence activity of 5 carriers significantly reduces as the result is shown; show that transcription factor C/EBP α inhibits the transcription of ACOX1 gene, to specify that transcription factor C/EBP α acts on the transcriptional control of ACOX1.In conjunction with bioinformatic analysis and site-directed mutagenesis technique, -1142~-1129 be C/EBP alpha transcriptional factor site as the result is shown. The present invention provides foundation for the research that the expression regulation of ACOX1 applies to livestock meat cytoplasmic inheritance improvement molecular regulation mechanism.
