abstract | The invention discloses a seawater nitrate reducing bacteria MCDA3-3, CGMCC NO.16934. The strain is a Gram-negative short bacillus without spores. Grow on 2216E medium at 25°C for 3 days, the colony is round, taupe and translucent, with moist surface, regular edges, no halo, central protrusion, diameter 0.1‑0.5mm, easy to pick. Produces a yellow transparent circle on selection media containing p-Nitro-N-acetanilide and powdered chitin. The invention also discloses a method for producing chitin deacetylase by using the bacterial strain MCDA3-3, and the optimum fermentation conditions are: 4% inoculum size, 30°C, 180rpm fermentation for 48h. The optimum temperature of chitin deacetylase produced by MCDA3‑3 is 30°C, and the optimum pH is 8.0. Metal ions such as Sr 2+ can promote the activity of chitin deacetylase. |
