abstract |
A method for increasing the color value of Monascus pigment in a fermentation broth is to complete the coding C-DNA in Monascus by using the Agrobacterium tumefaciens-mediated (ATMT) transformation T-DNA technology through a homologous recombination system that comes with Monascus. The ERG4 gene of 24(28) sterol reductase was knocked out to obtain a high-producing strain of Monascus pigment. The invention can significantly improve the production level of Monascus pigment in the fermentation broth. The results of liquid fermentation (10d) show that the extracellular color value and total color value of the starting strain are 14.57 U/mL and 106.54 U/mL respectively. The extracellular color value and total color value of the recombinant strain were 21.55U/mL and 112.78U/mL, respectively, and the extracellular color value and total color value were increased by 47.90% and 5.86%, respectively. |
