abstract | The invention provides a method for identifying the centromere chromosome in the middle of a patinopecten yessoensis, which selects three markers distributed on the SNP linkage map of the patinopecten yessoensis to screen a fosmid library by combining the genomic sequence information of the patinopecten yessoensis, the SNP linkage map information and the fosmid monoclonal decoding information, prepares probes with fluorescent markers by utilizing the clones containing SNP loci, then performs fluorescence in situ hybridization and co-hybridization by utilizing the probes, and realizes the specific identification of the centromere chromosome in the middle of the three pairs of patinopecten yessoensis according to the distribution of the fluorescent signals of the three specific clones containing SNP markers on the patinopecten yessoensis chromosome. The three markers provided by the invention realize the identification of three pairs of middle centromere chromosomes, and have important significance on the distinguishing and identification of the patinopecten yessoensis chromosomes, the construction of a cytogenetic map and the evolution analysis among the chromosomes from different species sources. |
