abstract | The embodiments of the present specification provide a method and application for ultra-low temperature removal of grape leafroll virus-3. The method comprises the following steps: taking a single stem segment of a grape branch, sterilizing and sterilizing, substituting and culturing on a primary medium, substituting the stem segment of axillary bud germination on a proliferation medium for cultivation, and taking the shoot tip containing 5-6 leaf primordiums. Planted on pre-medium, cultured in the dark; pre-medium-cultivated shoot tips were treated in loading solution, then placed on ice, and then made into droplets; cryoprotectant-coated shoot tips were placed in liquid nitrogen , then transferred to the unloading solution, thawed and unloaded; the thawed shoot tips were transplanted to recovery medium for dark cultivation to obtain plants without grape leaf roll virus-3. |
