abstract |
The present invention relates to the construction of a strain of Brucella rough with H gene of recombinant Peste des petits ruminants virus and a vaccine thereof. In the present invention, the attenuated strain RA343 of the rough-type Brucella bovis is used as the parental strain, and the sucrose suicide plasmid is used as the carrier, and the H gene of the Peste des petits ruminants virus containing a specific promoter sequence is codon-optimized and then inserted into the Brucella without trace. In the bacterial genome, a recombinant Brucella RA343-PPRV-H strain capable of highly expressing the recombinant PPR virus H gene was successfully constructed. The recombinant strain not only retains the rough-type characteristics of the original parent strain RA343 and has good immune protection against brucellosis (brucellosis), but also can produce specific neutralization against PPR after immunizing animals with the recombinant strain Antibodies to achieve immune protection against Peste des petits ruminants. The vaccine prepared by using the recombinant vaccine strain can simultaneously achieve dual immune protection against brucellosis and Peste des petits ruminants after immunizing animals. |
