abstract |
The invention discloses a method for inducing human pluripotent stem cells to differentiate into oocytes stagnated in meiosis II in vitro. The method is divided into three steps. First, the pluripotent stem cells with a cell density of 80% are replaced with PGC-m culture medium at 35-40°C and 3%-6% CO 2 for 10 days to obtain primordial germ cell-like cells; then transfer the formed primordial germ cell-like cells to PF-m medium, and at 35-40 Cultivated at 8% to 12% CO 2 for 5 days to obtain follicle-like structures; finally, the formed follicle-like structures were transferred to OLC-m medium and incubated for 10 to 15 days to obtain eggs that stopped at stage II of meiosis blast-like cells. The method of the present invention has high efficiency and short time, and the induced oocyte-like cells can exclude the first polar body and form secondary oocytes. This method provides a new means to study the formation and oogenesis of human female germ cells. |
