abstract | The invention discloses a composite agarose microsphere and a preparation method and application thereof, wherein the composite agarose microsphere comprises enzyme A, enzyme B, nitric oxide precursor derivatives and agarose, the enzyme A, the enzyme B and the nitric oxide precursor derivatives are wrapped in the agarose to form the composite agarose microsphere, the enzyme A is glucose oxidase or glucose-like oxidase, and the enzyme B is peroxidase or peroxidase-like enzyme. The preparation method comprises the steps of adding the enzyme A and the enzyme B into an agarose solution to form a water phase; dropwise adding the water phase into the organic phase, stirring and centrifuging, and collecting a centrifugal product; and adding nitric oxide precursor derivative into the centrifugation product for incubation to obtain the composite agarose microspheres. The composite agarose microspheres can stimulate the gel composite microspheres to controllably catalyze and generate nitric oxide by taking endogenous glucose as a reaction substrate, and have long drug action time and good biological safety. The preparation method is simple to operate and convenient to popularize and apply. The composite agarose microspheres can be applied to the preparation of medicaments for preventing and treating cardiovascular diseases. |
