abstract | The invention discloses a method for researching gene function of lentivirus transfected blastocyst trophoblast cells, which comprises the following steps: preparation of culture dish: placing 20.83uL of embryo culture solution into a centrifuge tube, adding 1uL of lentivirus, and blowing, beating and uniformly mixing; then, the embryo culture solution is dripped into a culture dish, 3mL of mineral oil is added, and the culture dish is placed in an incubator and balanced for 3-5 h. Lentivirus transfection of blastocysts: cleaning the blastocyst with the zona pellucida removed by using an embryo culture solution; the washed blastocysts are placed in a culture dish and transfected for 10-14 h; cleaning blastocysts with DPBS liquid, fixing with 4% PFA for 10-30min, and cleaning with DPBS; DAPI staining and tabletting. According to the invention, the interference on the expression of the target gene in the trophoblast cell is realized through lentivirus transfection, so that the problem that the gene in the trophoblast cell cannot be knocked down or overexpressed is effectively solved. |
