abstract The invention relates to a preparation and purification method of oligosaccharides of different regions of heparan sulfate/heparin (HS/Hp). The method uses commercially available heparin as a raw material, utilizes heparinase I to specifically enzymatically hydrolyze the high thiobiose containing 3 sulfite groups, precisely controls the enzymatic hydrolysis reaction conditions to achieve partial enzymatic hydrolysis, and obtains a high thiobiose containing high thiobiose. The sulfated region oligosaccharides also enriched the low sulfated oligosaccharide fragments containing NS and NAc structures in HS/Hp, thereby preparing a series of low sulfated region oligosaccharides with different structures. A series of low sulfated HS/Hp oligosaccharides were separated and purified by gel chromatography and ion exchange high performance liquid chromatography. The method for preparing oligosaccharides in different regions of HS/Hp established by the invention is fast, simple and low in cost, and provides an important sugar library sample for the structure and function research of HS/Hp, and also provides an important source for the development of new heparin drugs.
