abstract | The invention relates to a mass spectrum de novo sequencing method based on peptide fragment C-terminal selective enzyme labeling and application thereof. Under the catalytic action of amidase at the C-terminal of the peptide segment, the carboxyl at the C-terminal of the peptide segment is directly marked with small molecules with basic groups through amidation reaction or is firstly marked with unsaturated hydrocarbons with amino groups through amidation reaction, and is further connected with basic functional molecules through click chemical reaction of azide-alkynyl or sulfhydryl-alkene. The number of y ions in the secondary mass spectrum is increased by the basic molecules marked on the C terminal of the peptide fragment, and the response and fragmentation efficiency of the peptide fragment in the mass spectrum are improved, so that the sequencing of the peptide fragment is realized. |
