abstract | The invention discloses a large-scale qualitative method of acyl carnitine based on a mixed biological sample. According to different types of biological samples (blood, urine and tissue), acetonitrile/water is adopted as an extracting agent, vortex (blood, urine) and bead grinding (tissue) are adopted as an extracting method, supernatant fluid is obtained through centrifugation, vacuum freeze drying is carried out, acetonitrile/water is used for redissolving and mixing, and an ultra performance liquid chromatography-combined quadrupole ion trap mass spectrometer is adopted for analysis. And (4) screening out parent ions based on characteristic daughter ions and neutral loss obtained by the standard sample, and carrying out parallel reaction monitoring mode detection. The metabolic species obtained by detecting the sample by the method are rich, and the number of acyl carnitines which can be qualitatively obtained by one-time analysis is greatly increased. The method has the advantages of simple pretreatment, reliable qualitative determination, sufficient acquisition of acyl carnitine structure information and the like. |
