abstract | The invention provides a flagellin-fiber2 fusion protein, a preparation method and application thereof, wherein the fusion protein has fusion protein of avian adenovirus 4 fiber2 protein and salmonella typhimurium flagellin with high immunoprotection. The preparation method comprises the steps of fusing and cloning the artificially encoded flagellin-fiber2 gene into a pFastBacc-HA expression vector, transposing the gene to form recombinant Bacmid, then transfecting the recombinant Bacmid into Sf9 insect cells, expressing the fusion protein by using a baculovirus system, and identifying by indirect immunofluorescence IFA and Western blot. The period of obtaining the recombinant baculovirus by the system is short, and the flagellin-fiber2 fusion protein is used for immunizing SPF (specific pathogen free) chickens, so that the flagellin-fiber2 fusion protein expressed by the baculovirus system has high immune protection capability. |
