titleOfInvention | Method and kit for rapidly detecting horse-derived components in food |
abstract | The invention provides a rapid detection kit for horse-derived components in food, and relates to the technical field of biological species identification. The invention firstly screens out a horse-derived universal internal standard gene LOC106782588, the nucleotide sequence of which is shown in SEQ ID NO.1, the horse-derived universal internal standard gene is positioned on the 25 th chromosome, the copy number of the horse-derived universal internal standard gene is constant in horse species, allelic variation does not exist, and the horse-derived universal internal standard gene can be used as a target gene for identifying horse origin. An LAMP amplification primer is designed by taking the gene as a target sequence, and is subjected to constant-temperature amplification together with LAMP reaction liquid, and an LAMP reaction product is used for detecting the colloidal gold nucleic acid test strip. The LAMP reaction is combined with the colloidal gold nucleic acid test strip for detection to form a rapid detection kit, horse-derived components in food can be rapidly and sensitively detected, and the detection sensitivity can reach 0.2% (w/w). The kit has the advantages of simple use method, low cost, easy observation of reaction results and good specificity, and is very suitable for field real-time detection. |
