abstract The invention relates to the technical field of genetic engineering, and discloses a saffron-derived CCD2 mutant, a coding sequence and application thereof, and a recombinant yeast strain for producing crocetin. The CCD2 mutant provided by the invention has one or more than two of mutations at V120F, Y190K/A, R192V/F, E211A, E212A, T290V, K320A and S323F/A/T sites. The invention is modified aiming at the rate-limiting enzyme CCD2 in the synthesis path of the crocetin, realizes the direct synthesis of the crocetin from the beta-carotene by widening the substrate spectrum of the CCD2, simplifies and reconstructs the existing synthesis path of the crocetin, thereby solving the problem of generally low total yield of multi-step reaction; meanwhile, the efficiency of synthesizing the crocetin by catalyzing the original substrate zeaxanthin by the CCD2 is improved, and a yeast strain with high yield of the crocetin is obtained.
