In the present invention, the leuB gene in the genome of Bacillus thuringiensis Bt 4.0718 is successfully knocked out by using the I-SceI-mediated gene traceless knockout system to obtain a Bacillus thuringiensis mutant strain, and the ΔleuB mutant strain has obvious phenotype changes: In LB liquid medium, there were no mature spores, the amount of crystal protein synthesis increased, the mother cells did not split, and the crystal proteins were encapsulated to form micro-encapsulations; the insecticidal activity of the mutant strain against Plutella xylostella was higher than that of the wild strain, and the difference was significant. |
