abstract The invention provides a recombinant adenovirus vector construction and an adenovirus packaging method in which African swine fever EP153R and P54 genes are co-expressed, belonging to the technical field of genetic engineering. An EP153R gene and P54 gene over-expression adenovirus vector is based on the pShuttle-CMV eukaryotic expression vector, and CTLA4, EP153R and P54 genes are introduced into the multi-cloning site. The present invention also provides a recombinant adenovirus co-expressing EP153R and P54 genes, using the constructed pAD-Shuttle-CMV-CTLA4-EP153R-P54 vector to realize the adenovirus packaging process to obtain an adenovirus that can directly infect animals or eukaryotic cells , thereby realizing the purpose of co-expression of EP153R and P54 genes in eukaryotic cells, and laying a foundation for the study of adenovirus vaccines with co-expression of EP153R and P54 genes.
