abstract | The invention discloses a zero background cloning method of a gene with high GC content and application thereof. The invention successfully clones the complete CDS region and partial 5 'and 3' untranslated region sequences of the sheep Kiss1 gene with high GC content by screening and designing primers, optimizing an amplification system and applying an improved Slow-down PCR method and carrying out bioinformatics analysis. The nucleotide sequence of the sheep Kiss1 gene provided by the invention is shown as SEQ ID NO.3, and the amino acid sequence of the encoded protein is shown as SEQ ID NO. 4. The cloned sheep Kiss1 gene has wide application prospect in the aspects of inhibiting tumor metastasis and regulating and controlling the reproductive function of livestock. The invention provides a new research object for the research of the Kisspeptin related neuropeptides of the livestock, and the cloning method provided by the invention can provide theoretical reference for the cloning of other genes with high GC content. |
