abstract The invention discloses a separation and detection method of acetylcysteine enantiomer, which comprises the steps of carrying out derivatization reaction on N-acetyl-L-cysteine and an enantiomer thereof by using a derivatization reagent in an organic solvent to prepare a derivatization product of the N-acetyl-L-cysteine and the isomer thereof, and then carrying out separation and detection by using a normal-phase high performance liquid chromatography; the derivatization reagent adopts isocyanate. According to the separation and detection method of acetylcysteine enantiomer, a derivatization reagent reacts with N-acetyl-L-cysteine and the enantiomer thereof, and the mutual attraction between a fixed phase in a chromatographic column and a chiral substance to be separated is increased by adopting pre-column derivatization normal-phase high performance liquid chromatography according to a chiral separation mechanism, so that the selectivity of chiral separation is effectively improved. The method is simple and convenient to operate, short in time consumption and easy to popularize and use.
