abstract | The invention provides a method for constructing a melanoma nanobody library, which belongs to the technical field of bioengineering. The melanoma cells are cultured and then crushed, and 200ug/time of subcutaneously immunized alpaca four times; the sheep are collected one week after the fourth immunization. The camel anticoagulated blood, separated lymphocytes, extracted total RNA, and reverse transcribed into cDNA; using cDNA as the template, PCR amplification was performed with the first pair of primers, and the 700bp amplification product was recovered; using the 700bp amplification product as the template, the first The VHH fragments were amplified by PCR with two pairs of primers, and then ligated with pCANTAB5e after being digested twice with SfiI respectively; the obtained ligation products were electrotransformed into TG1 competent cells to obtain a melanoma nanobody library. The melanoma nanobody library constructed by the construction method of the present invention is an immune library with good characteristics and is suitable for specifically screening nanobodies related to melanoma growth, development, migration, proliferation and the like. |
