abstract | The invention discloses an in vitro verification method of banana fusarium oxysporum low-abundance milRNAs, which comprises the following steps: s1, designing a primer in the genome DNA of the fusarium oxysporum f.sp.cubense, amplifying a DNA sequence containing a candidate milRNA skeleton, and connecting the DNA sequence to an expression vector; s2, transforming the constructed vector into agrobacterium tumefaciens competent cells, and performing LB culture to obtain a bacterial solution; s3, infiltrating the leaves of the raw tobacco with the bacterial liquid obtained in the step S2, taking the leaves, carrying out corresponding small RNA probe hybridization detection, and judging whether the candidate milRNAs hairpin sequences can be processed into mature milRNAs. The method of the invention uses the Bunsen as a substitute plant, can effectively analyze the target and the function of the milRNAs, can perform high-flux analysis and verification on deep sequencing data, overcomes the defects in the current small RNA research, and has wide application prospect. |
