abstract | the invention discloses a mutant Fbn2 gene, which is different from a wild-type Fbn2 gene by 1 mutation, and the mutant Fbn2 gene causes the generation of trachea formation deficiency disease; the mutations are: c.2547T → A in Fbn2 gene; the invention also discloses a vector and a host cell containing the mutant Fbn2 gene and application thereof, and application of a reagent capable of improving the activity or/and expression quantity of the Fbn2 gene and an inhibitor of matrix metalloproteinase in preparing a medicament for treating the tracheogenesis deficiency. The invention constructs a novel Fbn2 gene mutant mouse (Fbn2) T2547A ) I.e., a mouse in which the nonsense mutation of Fbn2 results in decreased expression of Fbn2 protein and gene inactivation; it is also found that the inactivation of Fbn2 weakens the contraction of mouse trachea, the inactivation of Fbn2 leads to the disorder of arrangement of tracheal smooth muscle cells and the loss of polarity, the inactivation of Fbn2 causes the disorder of elastin formation and the reduction of fibrin expression, and the expression of Fbn2 and fibronectin in trachea of patients with trachea collapse is obviously reduced. |
