abstract The invention discloses a preparation method, system and recombinant bacmid of a recombinant adeno-associated virus (rAAV). The method comprises: (1) respectively preparing a shuttle plasmid and its corresponding recombinant bacmid containing a baculovirus genome; (2) combining an rAAV core expression element with a heterologous functional gene fragment with other functions necessary for the production of rAAV The expression cassette of the protein component is integrated to obtain a recombinant bacmid containing the recombinant baculovirus genome producing the rAAV; (3) transfection into a host cell line for culture. The system includes a shuttle plasmid and its corresponding recombinant bacmid containing a baculovirus genome. The recombinant bacmid contains at least one expression cassette of functional protein components required for the production of rAAV. The present invention has flexibility, compatibility and higher passage stability.
