abstract | The invention discloses circRNA related to MTX drug resistance of tumor cells and application thereof in reversing MTX drug resistance of tumor cells. The invention selects an HT-29MTX sensitive MTX drug-resistant cell line containing DMs as a research object, obtains circRNA highly expressed in MTX drug-resistant cells through second-generation sequencing, verification and optimization, and is named as circRNA7746, and further determines that the expression quantity of the circRNA7746 is positively correlated with the drug resistance degree of tumor cells to Methotrexate (MTX) by establishing an MTX treatment model of tumor-bearing mice and combining a PCR (polymerase chain reaction) and agarose gel electrophoresis method. The virus transfection method is utilized to respectively detect the influence of the interference circRNA7746 on the drug resistance of cells, the amplification degree and the amplification form of the DHFR gene and the micronuclear efflux, and the inhibition of the circRNA7746 is determined to promote the efflux of DMs and reverse the drug resistance of tumor cells to MTX. The invention provides a new target for tumor treatment and provides an effective technical means for effectively resisting MTX drug resistance. |
