abstract | The invention discloses a method for detecting hyriopsis cumingii lipid by using a modified sharp scalpel in real time through mass spectrometry, which comprises the following steps of: modifying the titanium base of the surface of the electrode of the riving knife so as to obtain a modified riving knife; pretreating hyriopsis cumingii tissues to obtain hyriopsis cumingii blocks B1; the method comprises the following steps of electrically cutting a hyriopsis cumingii block B1 by using a modified riving knife, ionizing lipid molecules in the hyriopsis cumingii block B1, and then atomizing into aerosol C1, wherein the aerosol C1 is mixed with nitrogen to form mixed gas C2 which is sucked into a mass spectrometer; the auxiliary solvent is 1.2% of leucine enkephalin propylene glycol solution, and data are collected in a negative ionization mode to obtain a hyriopsis cumingii mass spectrogram; aligning and denoising the hyriopsis cumingii mass spectrogram through MassLynx software, collecting mass spectrum peaks, and determining lipid molecules of a sample after comparing the mass-to-charge ratio information with a lipidomics database. The method can realize real-time mass spectrum detection and is suitable for lipid detection of hyriopsis cumingii. |
