abstract The present invention discloses a D-amino acid oxidase mutant and its application. The mutant is carried out by the 52nd, 54th, 58th, 213th and 335th amino acids shown in SEQ ID No.1. Obtained by single mutation or multiple mutations; wherein, glycine at position 52 is mutated to leucine, asparagine at position 54 is mutated to valine, phenylalanine at position 58 is mutated to glutamine, and position 213 is mutated to glutamine. The methionine was mutated to serine, and the serine at position 335 was mutated to glycine. In the present invention, the D-amino acid oxidase gene shown in SEQ ID No. 2 is mutated by using the site-directed saturation mutation technology to obtain a mutant whose enzyme activity and product conversion rate are much higher than those of the wild type, and the 4-(hydroxymethyl Product yield in phosphoryl)-2-carbonyl-butyric acid production process.
