abstract | The invention provides a construction and application of a novel bispecific CD20-CD19-CAR vector based on a humanized CD20 antibody and a CD19 antibody. Specifically, the invention provides a chimeric antigen receptor CAR comprising a CD 19-targeting scFv and a CD 20-targeting scFv, the CD 19-targeting scFv comprising the heavy chain variable region of SEQ ID No. 8 and the light chain variable region of SEQ ID No. 9, and the CD 20-targeting scFv comprising the heavy chain variable region of SEQ ID No. 10 and the light chain variable region of SEQ ID No. 11. CAR-T cells utilizing the bispecific CD20-CD19-CAR vector constructed in the present invention exhibited very good killing effect in vitro. |
