abstract Methods of generating nucleic acid libraries are provided. In certain aspects, the method comprises combining a target nucleic acid (eg, a 5' phosphorylated nucleic acid) and a pool of oligonucleotides. The oligonucleotides of the oligonucleotide pool may comprise complementary regions of different lengths and nucleotide sequences and complementary region recognition sequences. In this aspect, the combining is performed under the conditions that the oligonucleotides of the oligonucleotide pool hybridize to the nucleic acid (eg, a 5' phosphorylated nucleic acid) in the target nucleic acid, the Nucleic acids have overhang regions that are complementary in sequence and of corresponding length relative to the complementary region of the oligonucleotide. Compositions and kits useful, for example, in practicing the methods of the present disclosure are also provided.
