abstract | The invention discloses a method for determining activity of terminal deoxynuclease and application thereof. The method comprises the following steps: (1) designing a single-stranded DNA as a DNA probe; (2) respectively mixing TdT with DNA probe and CoCl at different final concentrations 2 Incubating dTTPs and buffer solution to obtain single-stranded probe DNA solution rich in poly-T sequence; then adding mercuric salt for incubation to obtain T-Hg II -a T-structure mediated double stranded DNA solution; adding nucleic acid dye to obtain mixed solution; finally, detecting the fluorescence intensity signal of the mixed solution, and drawing a detection curve according to the fluorescence intensity signal and the final concentration of TdT; (5) and replacing the TdT with the sample to be detected, repeating the steps, and then comparing the detection result with the detection curve to calculate the content of the TdT in the sample to be detected. The method has high sensitivity and good selectivity, and provides a new means for medical detection and biological research. |
