abstract | The invention discloses a method for constructing a high-throughput sequencing library of trace free RNA in body fluid. The method comprises the following steps: purifying body fluid sample exRNA by using a silicon matrix, dephosphorylating, adding polyA tail, and then carrying out reverse transcription on the tailing product by using template conversion and locked nucleic acid technology to obtain cDNA with a joint at the 5' end; then low-cycle PCR is carried out, and finally a high-throughput sequencing library is obtained. By applying template conversion and nucleic acid locking technologies, the invention deeply optimizes the problems of trace RNA in the construction process of a high-throughput sequencing library, and greatly reduces the library construction time and cost. By applying the technology of the invention, a library can be built for trace RNA less than 10ng in one day, then data with good quality can be obtained through high-throughput sequencing, and a plurality of existing RNA biomarkers related to diseases can be found through analysis. |
