abstract The present invention provides methods and compositions for detecting changes in the conformation of nucleic acid polymerases, the methods involving attaching a nucleic acid polymerase non-covalently attached to a single-walled carbon nanotube (SWNT) to a first nucleotide or a third a nucleotide analog is contacted with the template; and the conformationally changed nucleic acid polymerase is detected by measuring the first conductance change in the SWNT between the nucleic acid polymerase and the conformationally changed nucleic acid polymerase. The method is suitable for the sequencing of polynucleotides.
