abstract | The invention discloses a branched polyethyleneimine material containing thioketal bonds, and a preparation method and application thereof. The preparation method comprises the following steps: (1) stirring acetone and acid containing sulfhydryl group to react at room temperature; (2) after the reaction is finished, adding polyethyleneimine and an activating agent, dissolving in an organic reagent, stirring for reaction, and bonding chlorin e6 and carboxyl polyethylene glycol through amidation reaction to obtain the thioketal bond-containing active oxygen sensitive branched polyethyleneimine material. The branched polyethyleneimine material has good biocompatibility and degradability. The material carries siRNA, forms nano particles through self-assembly, generates a large amount of active oxygen to destroy an endosome structure under the excitation of a light source with specific wavelength, breaks down an active oxygen sensitive thioketal bond, disintegrates the particles, accelerates the escape of the endosome and releases the siRNA intracellularly, and has huge clinical application potential in the field of tumor treatment. |
