abstract The invention relates to a method for rooting populus diversifolia tissue culture seedlings outside bottles. The method comprises the following steps: removing the culture bottle of the populus diversifolia tissue culture seedlings with the seedling height of 6-8 cm from the culture room, closing the bottle and hardening the seedlings for 7-10 days; opening the bottle and hardening off the seedlings for 5 to 7 days; after the tissue culture seedlings of the populus diversifolia are subjected to induced rooting pretreatment by a liquid culture medium, transplanting the seedlings into a seedling culture tray containing a rooting medium, completely watering root fixing water after the seedlings are planted, and covering the seedling culture tray with a plastic film or a transparent container for moisturizing; spraying 2 times with an improved 1/2MS liquid culture medium every day in the early stage, wherein the temperature of a culture room is 25-30 ℃, the illumination intensity is 2000-2500 lux, and after indoor culture is carried out for 20-30 days, the rooting length of the populus diversifolia tissue culture seedlings reaches 5-7 cm; and finally, transplanting in a field. The invention has the advantages that: the tissue culture seedling ex-bottle rooting technology reduces the links of preparation, sterilization, inoculation and the like of a rooting culture medium, simplifies the production procedure, shortens the seedling culture time, and can shorten the overall seedling culture time by more than 20 days; the final rooting rate is 99.3%, and the survival rate is 98%.
