abstract The invention discloses a lipase mutant with improved thermal stability, a preparation method and application thereof, and belongs to the technical field of molecular biology. The amino acid sequence of the lipase mutant is such as SEQ ID NO.1 or SEQ ID NO.3 or SEQ ID NO.5 or SEQ ID NO.7 or SEQ ID NO.9 or SEQ ID NO.11 or SEQ ID NO.13 shown. According to the multi-sequence alignment and disulfide bond prediction results, the invention transforms the thermal stability of the Rhizopus oryzae lipase ROL, which greatly improves the thermal stability of the Rhizopus oryzae lipase ROL. The principle of thermal stability improvement is explained at the molecular level. The thermal stability of the lipase mutant provided by the invention is significantly improved, and combined with its high Sn-1,3 selectivity, it has more industrial application value.
