abstract | The invention discloses a method for analyzing the digestibility of a protein-polysaccharide complex based on a liquid chromatography-mass spectrometry technology. The method comprises the following steps: digesting the protein solution and the protein-polysaccharide complex solution in vitro in simulated gastric fluid; then, carrying out denaturation treatment on the digested protein and the protein-polysaccharide complex, removing low molecular weight polypeptide fragments by using an ultrafiltration tube, and further completely hydrolyzing large-fragment polypeptides trapped in the ultrafiltration tube by using endoprotease with a definite second enzyme cutting site except pepsin to obtain a plurality of polypeptide fragments; and finally, detecting the composition and abundance of the polypeptide by using a liquid chromatography-mass spectrometry technology. And determining the digestion rate of the pepsin according to the second protease cut and the polypeptide abundance of the pepsin cut peptide fragment. The combination sites of protein and polysaccharide are further deduced according to the high abundance of polypeptides in the group of protein-polysaccharide complexes in the two peptide fragments. |
